ESCHERICHIA COLI TEST STRIPS
PURPOSE OF USE
The Echerichia Coli Test strip is a specific test for the rapid identification of Escherichia coli using ß-glucuronidase activity.
METHOD
The following steps should be taken for the test:
1. Remove the strip tube and solution from the cooler and allow it to cool to room temperature.
2. Add 1-2 drops of E. Coli Test Reagent onto the test strip.
3. Pick up the suspected E. coli bacterial colony(s) with the help of a pipette and spread them on the reaction area of the test strip.
4. The result is determined by the colour formed in approximately 3 minutes.
RESULTS
Positive result: blue colour develops within 3 minutes
Negative result: pink colour develops within 3 minutes

OXIDASE TEST STRIBE /Cytochrome Oxidase Test in Microorganisms
Oxidase test strips are a rapid test tool for the determination of cytochrome oxidase enzyme. The strips are prepared by impregnation with reagents that rapidly change colour on contact with positive oxidase bacterial colonies.
Cytochrome oxidase enzyme is produced from various microorganisms belonging to the genera Pseudomonas spp., Neisseria spp. The oxidase test is a reaction used in the identification and differentiation of bacterial cultures. For a complete identification, it is recommended to perform additional tests with pure cultures.
METHOD OF USE
The following steps should be taken for the test:
1- The strip tube stored in the cooler is removed from the cooler and kept until it reaches room temperature.
2- A well-isolated colony is selected from the medium with a microscope.
3- The bacterial colony in the extract is carefully applied on the reaction part of the strip.
4- The colour formed in about 30 – 40 seconds is compared with the scale on the label on the strip tube.
REACTIVES
Each strip is impregnated with N,N-dimethyl-p-phenylenediamine and α-naphthol.
